Infection normally benign

Consequences: immuno-compromised/congenital infection

Retinochoroiditis

Important for children infected in utero

neontatal Swiss-Webster mice 

 

MATERIALS AND METHODS

primary neonatal infection & reinfection of neonatal infection

5 cysts (Test one)

4 weeks later both eyes

Day 3: inflammatory 

Bio-Plex mouse quantify cytokines and chemokines 

IFN-γ, IL-2, TNF-α, MCP-1, IL-6, IL-17A, IL-13, IL-10

 Tachyzoite (Test two)

Transcrption factors and cytokines, mRNA level



RESULTS TEST ONE

4 Week old mice

Primary: rapid ocular inflamation 

Cytokines IL-6 & IL-17A: lower in renfection

Chemokine MCP-1: lower in reinfection 

Th1 cytokine IFN- γ & Th2 type IL-13/IL-10: higher 

Th17 & Th1:IL-27 increase in mRNA

T-bet & GATA3: increase in renfection 

Most change occured in day 7

RESULTS TEST TWO

Primary mice died between day 3-7

Parasite multiplicaion faster in primary infection

No statistical difference

Cytokine levels IL-6 & IL-17: reinfection lower

IL-13/IL-10: still higher

T-bet levels higher in Reinfection

Lower IFN-γ


DISCUSSION

Less sever inflamation: reinfection 

IL-17: tissue distruction

reinfection: "natural" protection

Th1 & IFN-γ: necessary for control 

Rentinal cells more prepared in reinfection

Higher IL-10: control inflammation 

Th transcription  gives insight cell development 

Suppression of IL-17 requires counterbalance

Previous infection adds protection 

Protection not premeant
